Double-Labelled Physarum Amoeboflagellate
Tubulin and Actin
Confocal Microscopy of a Physarum polycephalum amoeba in the process of transforming into a flagellate swimming cell. Double labelling for tubulin (green, indirect immunofluorescence) and actin (red, rhodamine-phalloidin). Imaging carried out with the BioRad MRC600 Confocal Microscope, using the A1/A2 filter set (simultaneous capture of both FITC and Rhodamine images). Upward-pointing arrow indicates anterior flagellum. Downward-pointing arrow indicates apparent co-localization (red-green overlap yields yellow) of microtubules (MTA 5) and actin at the base of the ridge, a flattened motile projection found on transforming cells.
